The diurnal variation of nitric oxide (NO) emission fluxes from a Kandelia obovata and Avicennia marina mangrove wetland were studied in the Zhangjiang River Estuary Mangrove National Nature Reserve using a dynamic chamber-based technique and a chemiluminescent analyzer. Results from field experiments show that NO emission from K. obovata and A. marina sampling sites reached maximal values of 1.07 ng N m−2 s−1 and 1.23 ng N m−2 s−1, respectively after the night tide. Meanwhile NO emission maintained at a steady lower level in daytime for both wetland sites. In laboratory experiments, NO emission from the mangrove wetland soil samples treated with simulated tides in the darkness exhibited higher values than those in the light, therefore it seems that tides and darkness could increase NO emission from mangrove wetlands, while intensive light, high temperature, and dryness in the daytime decreased NO emission. Compared with K. obovata soil samples, the diurnal average NO emission rate of the A. marina site was significantly higher, which was closely related to relatively higher diurnal average CO2 emission rate, soil available nitrogen content and soil net nitrification rate of the A. marina site. Moreover, soil samples of the A. marina site were more responsive to simulated tides and the addition of nitrogen than those of the K. obovata site.  相似文献

Abstract

Abstract The MASONW (MACRO + SOILN + Watershed) model describing nitrogen leaching in watersheds was developed and tested. The model is based on the MACRO and SOILN models. The dual-porosity model MACRO simulates water flow on the field scale. The SOILN model describes turnover and leaching of nitrogen. Two main features of a watershed have been added into these two models: (a) the existence of a river system, and (b) variable thickness of the aeration zone within a watershed. Good agreement between the output of the MASONW model and observed data for water discharge and nitrate concentrations were achieved in the Odense watershed (496 km2) in Denmark.  相似文献

Storm events are major transporters of faecal microbial contaminants, but few studies have reported storm loads or concentration dynamics in relation to discharge or other pollutants, notably fine sediment. Episodically, high loads of faecal contamination during storm flows impact downstream uses of water bodies, particularly contact recreation and shellfish harvesting. We examined the storm dynamics of Escherichia coli, turbidity and discharge in the mixed land use Motueka catchment (2047 km2; 60% forest and 19% pasture) to gain insights into E. coli sources and transport. We also explored different approaches for calculating E. coli loads. Discharge and field turbidity were recorded continuously, and E. coli concentrations were sampled during events, over a 13‐month period near the mouth of the Motueka River. E. coli loads were estimated by interpolation, averaging estimators and by using linear regression with smearing correction of the log‐transformed variables: discharge, turbidity, and both turbidity and discharge. The annual E. coli load was dominated (~98%) by export during events. Comparison of monthly monitoring with the intensive storm monitoring campaign suggests that simple stratification of the sampling into storm and baseflow would greatly improve export estimates. E. coli peak concentrations always preceded discharge and turbidity peaks (which had similar timing). Turbidity can be a useful surrogate for faecal microbes in smaller catchments, but in the Motueka turbidity was no better for predicting E. coli concentration than discharge. Runoff from grazed pasture and direct deposition from livestock are probably the ultimate E. coli sources in the Motueka catchment. However, in‐channel stores seem to dominate E. coli dynamics during events and account for the typical feature of bacterial concentrations peaking ahead of discharge and turbidity. This study demonstrates the importance of storm events to faecal microbial loads and shows that E. coli concentration dynamics may contrast with those of turbidity. 通过CCK8法检测分离自深海一株深海耐压菌Shewanella piezotolerans WP3次生代谢产物的细胞毒活性,研究了温度、压力、噬菌体对WP3次生代谢产物细胞毒活性的影响.发现WP3次生代谢产物具有良好细胞毒活性.低温因素可提高其细胞毒活性,在100μg/cm^3的含量下,代谢产物对肿瘤细胞Bel7402、CNE2、A549、HT1080细胞毒活性均有提高,其中对A549细胞毒活性提高了1.3倍.在最适温度(20℃)情况下,高压可降低其细胞毒活性.20 MPa培养条件下,代谢产物对4株肿瘤细胞均未表现出明显活性,而常压培养条件下对SW 480具有93.3%的抑制率,对HT1080抑制率表现出87.5%,对CNE2抑制率是79.8%.在低温情况下,压力因素对其细胞毒活性无影响,噬菌体对细胞毒活性影响不大.HPLC指纹图谱分析发现不同温度条件下,其次生代谢产物指纹图谱有变化,结合细胞毒活性结果分析,为后期分离鉴定细胞毒活性化合物提供指导.  相似文献

为研究海水养殖对海湾沉积物的累积影响, 对大亚湾的大鹏澳养殖区沉积物柱状样中总氮、总磷和有机碳的含量及剖面分布特征进行了研究, 并探讨了海水养殖区沉积物中氮、磷的污染状况。结果表明, 网箱养殖区、贝类养殖区和对照区等3个区域柱状样中总氮的含量范围分别为638.2—3803.9mg.kg-1、740.9—2152.1mg.kg-1和343.2—471.9mg.kg-1, 总磷的含量范围分别为344.7—3233.9mg.kg-1、297.9—497.5mg.kg-1和650.2—1327.2 mg.kg-1, 有机碳的含量范围分别为0.96%—2.22%、0.87%—1.13%和0.69%—0.95%。该三个因子的含量均从底层至表层呈增加趋势, 但网箱养殖区上层增加幅度最为剧烈, 贝类区次之, 对照区变化幅度最小。所有柱状样中, 总氮含量均超标, 但网箱养殖区总氮污染最为严重, 贝类养殖区次之, 对照区最轻; 网箱养殖区的总磷在上层的不同深度超标, 而贝类养殖区及对照区柱状样的总磷含量均未超标。  相似文献

沼泽湿地土壤是氮的重要储库,发挥着源、汇和转化器的重要功能。湿地土壤中无机氮的物理运移不但影响着植物的养分供给状况,而且其对于湿地生态系统的结构、功能及健康状况等也有着深刻影响。综述了沼泽湿地土壤无机氮物理运移及影响因素的研究动态。当前湿地土壤无机氮物理运移的研究主要集中在硝态氮和铵态氮(特别是硝态氮)物理运移规律及部分影响因素(如水分条件、土壤物理性质等)的探讨上,缺乏无机氮物理运移的动力学、热力学机制与模型表征研究。鉴于当前研究中存在的问题,指出天然沼泽湿地是研究的薄弱点,其在今后应亟需加强的领域包括:①无机氮物理运移的驱动机制;②动力学、热力学与环境效应模型表征;③人类活动和全球变化对无机氮物理运移的影响。  相似文献

Concerns for microbial safety of surface water facilitate development of predictive models that estimate concentrations and total numbers of pathogen and indicator organisms leaving manure‐fertilized fields in overland flow during runoff events. Spatial variability of bacterial concentrations in applied manure introduces high uncertainty in the model predictions. The objective of this work was to evaluate the uncertainty in model predictions of the manure‐borne bacteria overland transport caused by limited information on the spatial distribution of bacteria in surface‐applied manure. Experiments were carried out at the ARS Beltsville experimental watershed site (OPE3) in Maryland. Dairy bovine manure was applied at a 59·3 t/ha rate on the 3·55 hectare experimental field. Faecal coliform (FC) concentrations in manure measured in 2004, 2005, 2007, and 2009 varied by 4 orders of magnitude each year. Both runoff volume and FC concentrations in runoff water were monitored using a runoff flume equipped with a refrigerated pump sampler. Two runoff events occurred before the manure was incorporated into the soil. A bacteria transport add‐on module simulator of transport with infiltration and runoff (STWIR) was linked with the event‐based kinematic runoff and erosion model (KINEROS2) to simulate convective‐dispersive overland transport, bacteria release from manure, reversible attachment–detachment to soil, and surface straining of infiltrating bacteria. The model was successfully calibrated with the field experiment data. Monte Carlo simulations were carried out to account for the spatial variation in FC in applied manure and uncertainty in the FC distribution in manure caused by the small number of samples. A tenfold and twofold variation in FC concentrations in the runoff were obtained within the 90% probability interval when initial FC spatial distributions in the manure were represented by 5 and 29 samples, respectively. 对个体重2.5~34.5g的牙鲆幼鱼在不同水温(12、17、22、27℃)下的耗氧率和氮排泄率进行了测试和分析。结果表明,牙鲆幼鱼的耗氧率和氮排泄率均随体重的增加而增加,其差异均达到极显著水平(P<0.01);牙鲆幼鱼的耗氧率随水温上升而增加,但仅27℃组与其他3个温度梯度组分别比较,出现差异极其显著(P<0.01),后3个温度梯度组之间的差异则不显著(P>0.05);牙鲆幼鱼的氮排泄率也随温度的提高而增加,其差异亦达到极显著水平(P<0.01)。作者认为,在牙鲆养殖生产过程中,养殖水体溶解氧应维持在3mg/L以上,养殖水温以不超过27℃为宜。  相似文献

从不同海区的海洋底质,生物样,水样中分离到480株海洋细菌,其中包括150株芽孢细菌和108株粪大肠杆菌,利用纸片法对其抑菌活性进行检测,结果表明:117株海洋细菌具有抑菌活性,占总分离菌株的24.4%;一般细菌,芽孢细菌,粪大肠杆菌的抑菌活性菌株分别占各供测菌株的17.1%,27.3%,35.2%;抑菌活性菌株的比例与样品来源密切相关,且各类海洋细菌的抗菌谱有很大的不同。  相似文献

To verify the hypothesis that the growth of phytoplankton in the Western Subarctic Gyre (WSG), which is located in the northwest subarctic Pacific, is suppressed by low iron (Fe) availability, an in situ Fe fertilization experiment was carried out in the summer of 2001. Changes over time in the abundance and community structure of phytoplankton were examined inside and outside an Fe patch using phytoplankton pigment markers analyzed by high-performance liquid chromatography (HPLC) and flow cytometry (FCM). In addition, the abundance of heterotrophic bacteria was also investigated by FCM. The chlorophyll a concentration was initially ca. 0.9 μg l−1 in the surface mixed layer where diatoms and chlorophyll b-containing green algae (prasinophytes and chlorophytes) were predominant in the chlorophyll biomass. After the iron enrichment, the chlorophyll a concentration increased up to 9.1 μg l−1 in the upper 10 m inside the Fe patch on Day 13. At the same time, the concentration of fucoxanthin (a diatom marker) increased 45-fold in the Fe patch, and diatoms accounted for a maximum 69% of the chlorophyll biomass. This result was consistent with a microscopic observation showing that the diatom Chaetoceros debilis had bloomed inside the Fe patch. However, chlorophyllide a concentrations also increased in the Fe patch with time, and reached a maximum of 2.2 μg l−1 at 5 m depth on Day 13, suggesting that a marked abundance of senescent algal cells existed at the end of the experiment. The concentration of peridinin (a dinoflagellate marker) also reached a maximum 24-fold, and dinoflagellates had contributed significantly (>15%) to the chlorophyll biomass inside the Fe patch by the end of the experiment. Concentrations of 19′-hexanoyloxyfucoxanthin (a prymnesiophyte marker), 19′-butanoyloxyfucoxanthin (a pelagophyte marker), and alloxanthin (a cryptophyte marker) were only incremented a few-fold increment inside the Fe patch. On the contrary, chlorophyll b concentration reduced to almost half of the initial level in the upper 10 m water column inside the Fe patch at the end of the experiment. A decrease with time in the abundance of eukaryotic ultraphytoplankton (<ca. 5 μm in size), in which chlorophyll b-containing green algae were possibly included was also observed by FCM. Overall, our results indicate that Fe supply can dramatically alter the abundance and community structure of phytoplankton in the WSG. On the other hand, cell density of heterotrophic bacteria inside the Fe patch was maximum at only ca. 1.5-fold higher than that outside the Fe patch. This indicates that heterotrophic bacteria abundance was little respondent to the Fe enrichment.  相似文献
